Cell therapies will only enable the treatment of diseases if an adequate number of therapeutic-grade cells are available. Thanks to their unlimited capacity for proliferation and differentiation, pluripotent embryonic stem cells (SCs) and their human-derived counterparts – induced pluripotent stem cells (iPSCs) – offer a means of meeting this requirement. Human iPSCs are particularly attractive, as they enable the preparation of autologous cells for a specific patient. The experiments included in this project will lead to the design and validation of techniques for the production of clinically relevant quantities of human progenitor cells of the vascular endothelium, pulmonary epithelium and smooth muscle cells, which will have a clear capacity to develop into mature, functional cells with therapeutic potential. This objective will be achieved by generating iPSCs from various types of human somatic cells and selecting the best ones based on their differentiation capacity and their safety in terms of genetic stability. These cells and the associated protocols will provide a source for the treatment of diseases characterised by abnormal vascularisation or lung epithelial function.
