Tau tubulin kinase 2 (TTBK2) is considered a key regulator of primary cilia formation, sensory organelles that play important roles both during embryonic development and in disease. However, we still have very little information available to form a comprehensive picture of what this kinase actually does in regulating cilia formation.
The research plan presented here focuses on analysing the molecular mechanisms and functional consequences of the initial steps in primary cilia formation, with particular emphasis on events that we hypothesise are regulated by TTBK2. Specifically, the project aims to contribute to clarifying what it is that makes the TTBK2 kinase so important for ciliogenesis by integrating ‘state-of-the-art’ proteomics and genetics (gene editing) with approaches from cell and molecular biology.
We believe that the research proposed here has the potential to significantly advance our understanding of the regulation of primary cilia formation.
