Cílem projektu je identifikovat, jakým způsobem enzym kasein kináza 1 (CK1) reguluje buněčné procesy v kontextu normální a nádorové buňky. S využitím nejmodernějších postupů proteomiky, strukturní biologie a sekvenování nové generace (i) identifikujeme, které molekulární a buněčné události jsou neredundatně kontrolovány CK1, (ii) popíšeme mechanismus fyziologické aktivace CK1 Wnt ligandy, (iii) na hlavním substrátu CK1 - proteinu Dishevelled - popíšeme na strukturní úrovni, jak CK1 aktivuje Wnt dráhy a (iv) na modelu chronické lymfocytární leukémie popíšeme jaké jsou důsledky inhibice CK1 pro patogenezi CLL. Tyto výsledky přiblíží klinickému využití CK1 inhibitory využité na MU a odhalí další molekulární události, které umožní cílit na procesy kontrolované CK1.
