The embryonic stem (ES) cells represent a potential source of material for cell therapies and regenerative medicine. Here, the gp130 receptor-mediated signalling pathway of the IL-6 cytokine family and their role in the regulation of ES cell differentiation will be studied. This pathway is known to support mouse ES cells in the undifferentiated stage. However, this pathway also regulates proliferation, differentiation, and apoptosis of many cells in the developing embryo and adult body. Two cardinal questions in this project will be investigated: a) The potential role and possible molecular mechanism of gp130 signalling for induction of specific lineages commitment of ES cells, and b) The fundamental mechanism-mediated shift between inhibition and induction of differentiation by the gp130 signal transduction pathway in mouse ES.
