Projekt se zabyva dvema enzymy,
avin reduktasou FerA a chinonreduktasou FerB, spojenymi s genero-
vanm nebo zvladanm oxidacnho stresu u bakterie Paracoccus denitricans, casto nazyvane

mitochondrie\. Bude izolovan gen pro FerA a pripraveny odpovdajc rekombinantn protein bude charak-
terizovan enzymologicky, krystalogracky a pomoc mstne rzene mutageneze. Bude stanoveno, zda FerA
pusob samostatne nebo v kombinaci s dalsmi proteiny. V pokusech in vitro bude kvantikovana produkce
superoxidu enzymem FerB a testovana hypoteza, ze FerB je schopen redukovat membranovy ubichinon. U divokeho kmene a mutantnch kmenu bez FerA resp. FerB nebo s jejich zvysenou produkc bude porov-
nan stupen peroxidace membranovych lipidu. Proteomicka analyza bunek vystavenych redoxnm stresorum
pomuze pri identikaci clenu redoxnho regulonu.
