It is thought that antigen-presenting cells (APSc), T-helper cells and cytotoxic T cells play crucial role in antitumour immune response. During the last decade dendritic cells (DSc), subset of APCs, has been identified to be the most important part for the effective antitumour response. DSc are the major APCs in activation of naive T cells "in vivo". The first studies started focusing on application of DCs in different malignacies, also in patients with multiple myeloma (MM). However, optimal protocol of DCs isolation, expansion amd pulsing with antigen is not yet known. In our hand, generation of DCs using a two-stage culture system whereby different cytokines are added at specified times during culture is used for expansion of DSc of MM patients. Fi rst optimal resorces of myeloid linaeage subtype of DSc will be evaluated in multiple myeloma patients. Further we will try to improve our current ptotocol of DSc cultivation including evaluation of optimal condition for pulsing DSc with antigen focused
