ALKBH5 is an m6A mRNA demethylase conserved across species, with roles in mRNA processing, export, localization, stability, and translation. Dysregulation of ALKBH5 impacts splicing, mRNA export, and stability, contributing to cancer progression. However, emerging evidence suggests that m6A abundance may be primarily regulated by exon-junction complex (EJC) deposition rather than ALKBH5 alone.
Our previous work identified ALKBH5 interactions with nuclear speckle proteins, splicing factors, and EJC components. Notably, its association with EJC core proteins and TREX export factors subbests a role in coupling splicing with nuclear export. This project aims to investigate the hypothesis that ALKBH5 functions as an auxiliary factor in mRNA export, ensuring quality control. Understanding ALKBH5´s molecular function is critical given ith implications in cances and development.
