Selective Plane Illumination Microscopy (SPIM) offers a promising advancement for assessing mechanical phenotypes of 2D and 3D cancer cell cultures. This project proposes a comprehensive platform integrating SPIM with flow stress induction (FSI) and mechanical compression for a high-throughput, cost-effective approach to cancer cell mechanophenotyping. By analyzing the viscoelastic properties of cells and subcellular structures, the system aims to unveil the role of mechanical forces in cancer cell behavior, advancing our understanding in drug discovery, developmental biology, and personalized medicine. By bypassing the limitations of techniques like Atomic Force Microscopy (AFM), proposed approach stands to significantly enrich phenotypic screening, facilitating the identification of novel drug candidates targeting cell migration and adherence. Given the pivotal role of cell mechanics in various biological processes, our approach extends beyond cancer research, potentially revolutionizing cell mechanics study across diverse applications.
